The Dynamic Organ Culture Incubator along with the Roller Inserts are used to optimally incubate tissue slices. The incubation space is enclosed and separated from the heating elements and controls. The incubator is maintained at 37◦C ± 1◦ and oxygenated through a port on the lid with 95% O2and 5% CO2. The temperature that the slice is exposed to can be monitored by placing a short thermometer into a water-filled glass scintillation vial which is closed with an open cap and a septum. This item can be purchased from Vitron. The O2/CO2 flow rate (0.5 – 1 liter/min) is controlled by a flow meter which is provided with the roller incubator. This roller culture incubator rotates the scintillation vial which in turn rotates the roller insert with slice so that all surfaces of the slice are exposed to the gaseous phase at 1 rpm. A total of 96 glass scintillation vials/roller inserts can fit into the incubator.
The slice is rotated in and out of the medium and oxygen atmosphere which is important in maintaining slice viability
The use of complete culture medium with fetal calf serum is recommended
It is recommended that the medium be changed every 24 hrs
Slice viability has been maintained in Dynamic Organ Culture for up to 7 days
